平凡大学生夜神月（洼田正孝 饰）偶然捡到写有“DEATH NOTE”的奇怪笔记本，起首部分还有如何使用的说明。完全没把它当回事儿夜神在上面写下了一直欺负他的高中同学的名字，谁知对方竟然真的在预定时间意外死亡。事后他又利用死亡笔记救下被歹徒劫持的父亲，从而确认了这个笔记本的真实性。与此同时，模样恐怖的死神琉克（福岛润 配音）出现在青年面前，声称夜神已成为死亡笔记的主人，如何使用由他自己决定。在此之后，夜神利用笔记本杀死数十个罪大恶极之人，由此被网民奉为神明基拉。可是他的动向很快被天才少年L（山崎贤人 饰）查明，L宣称要将基拉绳之于法。
死亡笔记的残酷刚刚开始，更可怕的对决即将到来……
