Set in China in the 1930s, the film is about the unsettling relationship between three characters, each involved with a performance of the opera Fleeing by Night in a local theatre.

多年以前，在中国某个偏远的地方，一个盲童跟随师傅学琴，师傅告诉他有一道药方可以治愈他的双眼令他重见天日，而这道药方必须在他弹断一千根弦后方能从琴匣里取出来，否则就没有效用。盲童在师傅死后，弹断了一根又一根的琴弦，不顾人生其他。在岁月的磨砺下，小少年成了老琴师，并被村民们尊称为“神神”（刘仲元饰）。村内出现任何纠纷，大家总爱问神神排解。
神神有一个瞎眼徒弟石头（黄磊饰），他与村女兰秀（许晴饰）偷情被村民发现，遭到痛殴，兰秀因此愤然绝去。黄天不负苦心人，神神终于弹断了千根弦，只为师傅说那时方可得到药方、见到光明，然而事情却有着意想不到的结局。

在城市的破败角落，脑顶光秃秃的重案组老警察入职三十年，虽然破了许多大案要案，满身都是伤痕弹孔，但是他的家庭情况却一团糟。老婆和他离婚，儿子嫌他臭。与老警察相对，青年私家侦探波比（黄磊 配音）如日中天，扶摇直上，FBI请他讲课，相关动画片热播，波比俨然成为福尔摩斯一般的存在。在某起密室杀人案后，老警察不得已请来波比调查。波比为什么会这么厉害，这一切似乎都与他的妈妈麦太（吴君如 配音）有关。在崇拜他的小孩子们的追问下，波比的思绪回到孩童时代，麦兜（张正中 配音）和妈妈共处的那个快乐无忧的童年，一切美好都早已注定……
